چکیده

Identifying and characterizing the biochemical variation in plant tissues is an important task many research fields. Small spectral differences of cell wall that are caused by genetic or environmental influences may be superimposed individual as well a microscopic heterogeneity molecular composition structure different histological substructures. A set 56 samples from Cucumis sativus (cucumber) plants, comprising total ~168,000 spectra tissue sections leaf, stem, roots was investigated Raman microspectroscopic mapping excited at 532 nm. multivariate analysis carried out order to assess with respect origin tissue, (cell wall) substructures, possibility discriminate obtained individuals had been subjected two conditions during growth. Combining results principal component (PCA) based classification original spatial information maps 23 leaf xylem, found for four also includes discrimination grown presence absence additional silicic acid irrigation water plants. The data point contribution carotenoids, hydroxycinnamic acids spectra. give new insight into chemical useful elucidating processes associated biomineralization vibrational spectroscopy.

2D-PCR: a method of mapping DNA in tissue sections.

A novel approach was developed for mapping the location of target DNA in tissue sections. The method combines a high-density, multi-well plate with an innovative single-tube procedure to directly extract, amplify, and detect the DNA in parallel while maintaining the two-dimensional (2D) architecture of the tissue. A 2D map of the gene glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was created...

Raman Imaging of Plant Cell Walls in Sections of Cucumis sativus

Raman microspectra combine information on chemical composition of plant tissues with spatial information. The contributions from the building blocks of the cell walls in the Raman spectra of plant tissues can vary in the microscopic sub-structures of the tissue. Here, we discuss the analysis of 55 Raman maps of root, stem, and leaf tissues of Cucumis sativus, using different spectral contributi...

Characterization of hyaluronic acid on tissue sections with hyaluronectin.

An affinity immunological procedure for hyaluronic acid detection on tissue sections is described. This new, sensitive, and specific technique is based on the high affinity of hyaluronectin for hyaluronic acid, utilizing anti-hyaluronectin-hyaluronectin immune complexes. Elimination of binding when the reagent was supplemented with hyaluronic acid or when Streptomyces hyaluronidase-digested tis...

A cryohistological protocol for preparation of large plant tissue sections for screening intracellular fluorescent protein expression.

In this study, we have developed a robust cryohistological method that allows imaging of virtually any type of plant cell or tissue while preserving fluorescent protein signals and maintaining excellent cellular and subcellular morphology. This method involves modified fixation of plant tissues (i.e., leaves, stems, and petioles), infiltration in a sucrose gradient, freezing, and collection of ...

Effects of tissue fixation on Raman spectroscopic characterization of retina

Raman spectroscopy is a non-invasive and non-destructive tool that has been widely applied in Agricultural and Bio-system Engineering field. It provides a unique “fingerprint” to different chemical, and structural, information at molecular level. Acquisition of Raman spectrum requires a relatively short collecting time and minimum sample preparation, which makes Raman spectroscopy an attractive...
